Crepidiastrum denticulatum (CD) is a well-known, traditionally consumed vegetable in Korea, which was recently reported to have bioactive compounds with detoxification and antioxidant properties. Ischemia-reperfusion injury (IRI) is a major critical event that commonly occurs after liver transplantation. We investigated whether CD protects against IR-induced acute liver injury in mice.
Partial (70%) hepatic IRI was induced in male C57BL/6 mice by portal triad pedicle occlusion for 45 min followed by reperfusion for 6 h. CD (75 mg/kg BW, oral) was administered 5 days before the IRI.
Treatment with CD significantly decreased serum sALT, sAST and LDH as well as histological changes . CD prevented hepatic GSH depletion and increased MDA levels. Western blotting indicated that CD significantly increased the levels of Atg7, LC3-I/II, attenuated PARP and caspase3. The expression of the Toll-like receptor 4, iNOS and NFkB were decreased in the CD treatment group .
CD improved the acute hepatic IRI by reducing oxidative damage, apoptosis, inflammation and inducing autophagy. These findings suggest that CD is a promising agent against acute IR-induced hepatic damage.
