Prognostic Value and Biological Significances of Five Long Non-Coding RNAs in Hepatocellular Carcinoma

Objectives Increasing evidence suggests that long non-coding RNA (lncRNA) plays a vital role in metastasis and development in hepatocellular carcinoma (HCC). Tumor progression severely affect the prognosis of HCC patients. We aim to explore the prognostic value and biological significance of lncRNAs in HCC.

Methods LncRNA expression profiles and clinical data of a TCGA HCC cohort of 364 patients were analyzed. Multivariate Cox proportional regression was performed to construct lncRNA-based risk score and overall survival (OS) prediction nomogram. KEGG pathway and GO pathway functional enrichment analysis was performed. A competing endogenous RNAs (ceRNAs) network was constructed by miRcode. Digoxin-labeled hybridization(CISH) was used to detect the lncRNAs' expression level in 95 pairs of HCC and para-carcinoma tissues from our center.

Results Five lncRNAs were identified with significant statistics difference (P<0.001). Risk score= (10.0542[times]CTD2116N20.1)+(0.0223[times]AC012074.2)+(-0.0004[times]RP11-538D16.2)+(0.0136[times]LINC00501)+ (-0.035[times]RP11-136I14.5). An OS prediction nomogram was made with the C-index of 0.701. Co-expression network results indicate that four of these prognostic lncRNAs are regulators of proteins in HCC-derived exosomes. CeRNAs network reveals possible mechanisms of RP11-538D16.2 and CTD2116N20.1 regulating the expression of proteins in exosomes. CISH results shows those two lncRNAs are significantly overexpressed in tumor tissures.

Conclusion Our findings provide an effective prognostic model to predict the survival rate of HCC patients. RP11-538D16.2 and CTD2116N20.1 were highlighted and their ceRNAs network was constructed.
