Efficacy of Photobiomodulation Therapy Combined with Short-Term Immunosuppressants Prolonged Allotransplant Survival in a Rodent Hind-Limb Model

*Purpose: Photobiomodulation has been applied for accelerating the skeletal muscle repair process. Our formal study demonstrated vitamin D could be involved in allograft survival and tolerance induction. This study we want to investigate whether photobiomodulation combined with short-term immunosuppressant could modulate allograft rejection.
*Methods: Orthotopic hind-limb transplants from Brown-Norway (BN) to Lewis (LEW) rats were performed (day 0). Group 1 did not receive any treatment, as rejection group. Group 2 received anti-lymphocyte serum (ALS; -4, +1) combined with CsA (days 0-21) following VCA. Group 3 received recipient ASCs (rASC) harvested from LEW ASC following VCA (2-3×106/ time, days +7, +14, +21). Group 4 treated with full spectrum photobiomodulation therapy for 8 hrs per day.
*Results: The results revealed treatment with photobiomodulation therapy combined with short-term immunosuppressant resulted in a statistically significant increase in allograft survival. The H&E staining performed on the allo-skin and muscle biopsy samples exhibited lowest degree of rejection and minimal mononuclear cell infiltration in the donor skin or muscle tissue in photobiomodulation therapy group as compared to that in controls. Flow cytometric analysis in photobiomodulation therapy group revealed a significant increase in the percentage of CD4+/CD25+/Foxp3+ T cell population in the recipient peripheral blood.
*Conclusions: Combination of photobiomodulation therapy and short-term immunosuppressant could prolong VCA survival. The bio-mechanisms of photobiomodulation therapy need further elucidated in the future.
