Permanent hypoparathyroidism (PH) is a serious clinical condition and unique treatment option is parathyroid allotransplantation (PA). The most important obstacle for PA is an immune response to transplanted parathyroid cells. Microencapsulation techniques are used for various cell type allotransplantations to protect transplanted cells from host immune system via biocompatible polymers. In this study we xenotransplanted microencapsulated human parathyroid cells to the intraperitoneal cavity of rats and with long-term results. 30 Wistar albino female rats (4 months old, 200-220gr) were divided into three groups. Parathyroidectomy and thyroidectomy performed for all subjects with small neck incision before transplantations. In group 1, microencapsulated human parathyroid cells with ultrapure alginate were transplanted (n:10), in group 2, empty microcapsules were transplanted (n:10), in group 3 human parathyroid cells transplanted without microcapsules (n:10). Blood samples were collected for 12 weeks and parathormone was determined (Figure 1). Our data suggest that long-term survival and function of microencapsulated parathyroid cell transplantation may solve the major drawbacks of immune rejection associated with parathyroid allotransplantation survival with this improved efficacy. Moreover, ultrapure alginate based microencapsulated parathyroid transplantation may be an effective and promising technique for the treatment of PH.
