Aims: The incidence of fungal infection in post-transplanted patients is 13%-17%, which seriously effecting perioperative survival and increasing their economic burden. We wanted to find out novel biomarkers for the early and fast diagnosis of fungal infection.

Methods: Mice were infected with fungus (e.g. Candida) on the 7th day after skin transplantation. Peripheral blood was collected and screened by gene chips. Differentially expressed miRNAs and lncRNAs were humanization compared, verified by qRT-PCR and confirmed in the 1043 blood samples of post-transplanted patients.

Results:

After gene chips screening, humanized compare and qRT-PCR verification, differentially expressed 20 miRNAs and 16 lncRNAs in peripheral blood were discovered. Serum level of miR-215 and miR-let-7c increased significantly in patients with fungal infection (P<0.001), while the expression of miR-154, miR-193a, NR_036506.1 and NR_027669.1 in peripheral blood mononuclear cells decreased notably (P<0.001). Moreover, similar changes of these biomarkers were observed in patients infected with Mucor, Aspergillus or Cryptococcus in pulmonary, abdominal cavity or digestive tract.

Conclusions: The level of miR-215 and miR-let-7c in serum and miR-154, miR-193a, NR_036506.1 and NR_027669.1 in peripheral blood mononuclear cells may be potential and helpful biomarkers for fungal infection in post-transplanted patients.
