Aims: Mortality of CMV infection in post-transplanted patients was quite high, which severely influencing the long-term survival of recipients. For giving them early treatment and improving their prognosis, we aimed to discover a novel diagnostic method to enhance the early diagnosis rate of CMV infection.
Methods: A murine model of CMV infection after skin transplantation was established.Peripheral blood was collected at a certain time point and screened by gene microarrays. Differentially expressed microRNAs (miRNAs) and long non-coding RNAs (lncRNAs) were homologous analyzed with those of human and verified by qRT-PCR. Finally, we confirmed the potential biomarkers in the blood sample of post-transplanted patients.
Results: After gene chips screening, humanized comparation and qRT-PCR verification, differentially expressed 13 miRNAs and 7 lncRNAs in peripheral blood were discovered. Expression of miR-29b, miR-185 and NR_073415.2 in peripheral blood mononuclear cells were significantly decreased in patients with cytomegalovirus infection (P<0.001) during the early stage and kept downregulating for more than 21 days.
Conclusions: Circulating miR-29b, miR-185 and NR_073415.2 were decreased in post-transplanted patients with cytomegalovirus infection, and they may represent candidate biomarkers for early diagnosis.
